Salcaprozate Sodium (SNAC)
Salcaprozate Sodium (SNAC) (Sodium 8-[(2-hydroxybenzoyl)amino]octanoate) is a type of permeation enhancer used primarily as a specialized pharmaceutical excipient to increase the oral bioavailability of small to large molecule APIs that otherwise cannot effectively be administered orally.
SNAC increases drug absorption through several mechanisms. Unlike many other enhancers that only temporarily open tight junctions between cells, SNAC typically acts as a "chaperone". It has the potential to non-covalently bind to medication, acting as a transport vehicle. SNAC is well studied in its numerous oral penetration enhancing benefits, such as its ability to raise local pH and protect APIs from stomach acids and enzymes that would otherwise destroy pH labile drugs and peptide medications.
SNAC is a well know permeation enhancer used in several FDA-approved oral peptide formulations including the FDA-approved GLP-1 receptor agonist, semaglutide. Salcaprozate Sodium (SNAC) holds Generally Recognized as Safe (GRAS) status.
